Tattooing goes way back in human history.

We've been inking ourselves for thousands of years.

And yet, exactly how the ink sticks around has remained somewhat of a mystery.

The rough idea was that either some skin cells or immune cells absorb ink particles, and

then basically never die.

Unless it's been a long time or they're blasted apart by lasers.

But a new study in The Journal of Experimental Medicine is here to set the record straight:

it is immune cells that suck up the ink.

But they die all the time, and new ones take over the ink hoarding.

At least, that's what research in mice indicates.

When you're getting tattooed, tiny needles poke holes that let the ink into your second

layer of skin, called the dermis.

Previous studies have looked at tattooed skin under a microscope.

But when you're looking at a bunch of ink-filled blobs, it's tough to learn specifics — like

which cells suck up the ink, how long they live, or what happens to the ink when they

die.

So this team took a different approach.

They used genetically modified mice that let the researchers flip a genetic switch and

kill off macrophages, the immune cells that travel around your body gobbling up foreign

substances and other debris.

And they used machines that can sort cells by really specific characteristics that a

microscope can't see, like what proteins are on the outside.

That way, they could dig deep into which cells were there, and which had ink in them.

Through these experiments, they discovered that a unique kind of dermal macrophage is

probably involved.

Specifically, it's a kind of melanophage, which are cells that ingest melanin, the pigment

responsible for tanning and variations in skin color.

Unlike other kinds of macrophages, they don't carry their contents away to be destroyed—they

stay put and hold onto that pigment.

And when the researchers gave their mice green tail tattoos, they found that melanophages

were picking up the ink.

When the researchers killed these cells off, the ink hung out in the goo between cells.

Then, as the melanophages were replaced with new ones from the blood, they sucked up the

same ink.

To see if this kind of turnover is normal, they grafted tattooed skin from one mouse

to another.

And sure enough, while all the ink-containing cells were from the graft donor at the start,

after about 6 weeks, they were mostly from the recipient instead.

Of course, this was work done in mice, so it may not perfectly model human tattooing.

But our skin does contain similar melanophages.

And the findings could explain why laser tattoo removal is so tricky.

The goal is to use light to break the ink pigments into smaller chunks and kill the

cells holding them.

But before all the ink is whisked away by your immune system, nearby melanophages might

absorb it again, helping your tattoo stick around — whether you want it or not.

Genetic engineering is a bit of a theme this week, from modified mice to plants.

Crops are among the thirstiest organisms on Earth, guzzling up to 90% of the world's

available fresh water.

And, like, we can't just stop growing them, because we need to eat.

Not to mention, we're staring down a future of widespread droughts thanks to climate change.

So this huge water problem is one that genetic engineers have been trying to solve.

And this week we may have taken one step forward.

A new study published in Nature Communications describes a single gene that can be manipulated

to make plants more tolerant to drought.

The new finding comes from an international research project called Realizing Increased

Photosynthetic Efficiency or RIPE.

It's a huge collaboration by seven institutions and dozens of scientists.

They're studying the remaining mysteries of photosynthesis, the process by which plants

use light energy to produce sugars and oxygen from carbon dioxide and water.

And now, they've discovered that increasing the amount of one protein—called Photosystem

II Subunit S or PsbS—creates water-conserving plants.

PsbS is found in all crop plants because it's one of about 30 proteins in Photosystem II,

the machinery that captures light energy.

When plants absorb more light than they need, PsbS helps dissipate some of the absorbed

energy as heat.

So when there's a whole bunch of PsbS, more light energy is diverted.

The plants are basically tricked, so they aren't getting ready to photosynthesize

as much, and need less of the other ingredients.

Plants take in carbon dioxide through little pores on their leaves and stems called stomata,

which can open or close to let in different amounts.

And these pores let water evaporate as a side effect.

But with more PsbS tricking the plant, stomata will stay partially closed, preventing some

CO2 uptake /and/ water loss.

To figure all this out, the researchers took tobacco plants and engineered them to produce

more or less PsbS.

They tested four overproducing and two underproducing varieties against non-engineered plants by

growing them in fields, where there's a little less control than greenhouses or labs.

And they found that the PsbS overproducers lost an average of 25% less water than the

non-engineered tobacco plants.

Even with partially closed stomata, the engineered plants had plenty of CO2 because there's

a lot more of it in the atmosphere than there used to be… thanks to us.

The main downside the researchers found was that half of their engineered plants were

less productive than the normal ones — they were shorter, had less mass, or had smaller

or fewer leaves.

But half weren't, and they weren't entirely sure why.

And they note that their experiment was done with lots of available water.

But if they'd simulated a drought, they expect the genetically modified tobacco would

perform even better.

Now, they aren't going to unleash these plants on the world anytime soon.

This is mostly just proof-of-concept that manipulating one gene found in a lot of plants

can make water conservation possible.

In the future, the RIPE researchers hope to engineer plants that can thrive in dry, nutrient-poor

soils.

But until these plants are made and tested there's no way to know how successful or

revolutionary they'll be.

And that's how science works.

It takes a lot of clever people working together to learn, experiment, and solve problems — both

in the classroom and out in the field.

So what's in the box?

According to the hang tag, the original MSRP for this watch was $24.99.

I picked this up on eBay as a department store return for much less.

...and it seems in perfect shape!

It has an unsigned metal buckle and keeper.

The watch is much dressier than most digital watches with a brushed stainless steel top

bezel, and flush side buttons.

The display sits at a tilt towards the wearer, which is a nice touch.

The large digits make this watch easy to read, but I wish the crystal was more anti-refelective,

as it can be hard to read under bright light.

The watch has the standard set of basic digital features.

Time, stopwatch, alarm, countdown timer,

as well as a second time zone.

The stopwatch is pretty cool, a blinking running man appears on the dial, and the days of the

week make an interesting chase pattern at the top of the display!

The countdown timer is interesting also, you can set it for any number of hours/minutes

and it starts to beep when there are only ten seconds left.

Check it out!

The back of the case gives the model and lets us know this watch has 100 meters of water

resistance.

Made in China... of course...

The rubber strap is 27mm wide at the springbars, and as you can see from the wrist shot, it's

definately long enough to accomodate my large wrist.

The watch features an electro-luminescent display, which means it looks fantastic at

night.

So yeah, overall I'm very pleased with this watch.

I think with the brushed stainless case and minimalist

button design, you could almost call this a dress watch.

Unboxing the Gigabyte Sabre 15 - Budget Video Editing Laptop That's Worth It! - Duration: 7:06.

- two minutes and nine seconds it's taking me 15 goodness morning everyone

it's Dennis here and today I'll be unboxing my new laptop the Gigabyte Sabre 15

I'm struggling with a lot of things with my computer my computer here is five and

a half years old and I think you know what it's it's about time that I updates

my computer so decided to pick a laptop and bukka gigabytes Avery fifteen now

probably look for like really good deals over the last couple of weeks leading up

to Christmas and you know just checking through websites comparing prices

compare specs and everything but I came to a point where it's like I can't even

make a decision what do I get the XPS 15 which I know is a really good laptop but

it's way above my budget or do I get the Acer predator which again it's a gaming

laptop yes so opted out with the gigabyte I checked out the reviews I've

checked out you know Dave 2d and a couple of guys from YouTube and they

said it's alright it's a budget laptop you know it has a 1050 TI graphics card

4 gig just good enough for video editing boredom for me is a processor it's an i7

7700 hg quad-core 2.8 gigahertz i think that's very very good for video editing

and it's a 15 inch laptop it has a 256 SSD and one terabyte which is enough for

me to put in videos you know my drone videos everything and jump in there and

it's portable sort of lights about 2.8 kilograms you don't the difference this

one with the Dell XPS 15 is the four Cape animals team basil it's very light

the SSD I think I believe is a lot faster with the Dell and yeah so I mean

there's a few hundred laptops in there I put I could list it down below so the

some of the laptop spec stuff looked at and tested out and so no camera computer

shops you never know that might help you

choose your video editing laptop who knows let's open it up shall we

Wow as RGB keyboard lighting HDMI USB and your jacks cool yeah so yeah that's

the laptop I'm just gonna put a connector charge it off now just start

installing some of the programs that I made and hopefully in an instant you'll

see some of the video rendering times of the footage and hopefully they'll give

you an idea of how this laughs that works so yeah I'm really excited like I

said I've struggled to edit with my current computer especially once I

started putting color grading color correcting and everything and I'll see

you in a bit finally installed Adobe Premiere Perl into my new laptop and I

just want to show you in comparison of the issues I've been having on my

current computer versus the gigabyte laptop that I have now and I have to say

I can really see the difference the laptop it's a lot faster it can color

great they can do 4k videos the struggle is very real at the moment with my

current computer they're very patient you know video editing using this

computer but I came to a point you know what I really need a new editing machine

and I need something portable and lie and say like I need something portable I

really don't have much money to spend so I currently buy those high-end ones so

my initial thoughts on barring the gigabyte pretty happy so I'm going to

show you you know the Christmas video that I've done I'm using a samsung sa

it's 4k file and without any sort of color grading or anything or you know

without any color correction it plays smoothly but once I start putting in

color correction and color grading whatever it just won't render in real

time or won't play Tom sell and I'll show it to you okay so

this is the 4k file the Christmas I'm just gonna press play so it's play

smoothly so far eyes without any color grading long to color collection it's

working fine okay so I'll just put a screen if it's the blurring but yeah

okay now you can see it's slowing down already

plus it for a while okay now well let's just say a drag blue cold there see it's

choppy it's choppy it's struggling let's say if I want change the saturation too

high vibrance too high sharpen too high if I change all the

sharpen hi see it stopped yeah it's skipping really struggling in

there with my computer there and it's just shopping it skips it doesn't and

that's just that's a 4k flower even for ten eight even a 1080p file actually

let's try that again how about some adjustment layer this is a 1080p file

sorry apologies significant in a show t oh okay okay so you can still see it you

still fine now if I hit this and I'll let say a blue moon to change the shop

into 1000 there you go again this is 1080p file that's struggling let me show

you how the Gigabyte works now let's continue I dropped in a 4k file I've got

a map a drone video here and then I've got the three Samsung sa files over here

as well you know the Christmas files and if I press go play for my mouse it's

full and it's fit it's what it's fine yellow bars here so it's not too bad now

but change the look say to blue Cole BAM it's still playing to change the sharpen

vibrance saturation - hi so you see here bump them up and it's

still playing so it's still on your level which is not

too bad okay this one again there's nothing here

change the gold raj HT are still playing bump up the saturation vibrance let's

say we play around with the cards yeah it's fine let me just try it again see

playing with the curve it's still playing do it so with my PC

it'll struggle with that one now if I'm gonna try and export this see saying six

six minutes just to export a three and a half file you know with color grading

color correction and those lots just so happy that I've got a laptop now that I
