Hi! Welcome to another Christmas baking.

Now I'm going to prepare triangles that are not exactly baked.

They're from almonds and

sugar... that's all.

140g of sugar that we will

boil with two spoons of water.

I'm very curious about that...

Very curious what that means.

Just had to check if I'm still filming.

Sometimes it happens that I'm not...

What will I boil it in?

I guess in this one.

Two spoons of water.

There's no spoon!!

Where are all the spoons?

Confusion.

And when it's in this phase,

we mix it with the ground almonds.

And add the rum.

We have 120g of ground almonds.

You're supposed to put two spoons of rum in it.

Which of course you don't have to do if you have a problem with rum.

Or alcohol itself.

But it will give it a nice smell.

Not that it would taste of rum or have much alcohol in it.

But it will create the aroma.

And mix it all together.

So here I've got melted chocolate.

Which I'm going to use to

coat the triangles.

But firstly we have to roll the dough which has cooled down.

We can use a little bit of powdered sugar

to help it not stick to the desk.

Also you can add a bit into the mix,

I added a little bit.

Maybe a handful.

Roll it into a rectangular shape...

cNMA web server demo [with audio] - Duration: 7:58.

Hello, I am Yuanfei from Dr Shen's lab.

Today, I will show you a quick demo of our cNMA web server

which serves as an efficient and accurate tool to predict protein conformational changes

using our newly proposed

framework of contact-specific Normal Mode Analysis

called cNMA and machine learning approaches

As you can see, we have two pages here: home page and tutorial page

Jobs will be submitted on home page also

Result files will be presented here

You can find more explanations and instructions on tutorial page

We provide two analysis modes here: one protein mode is for single protein in unbound state

Two proteins mode is for unbound and homology docking

Now I'm gonna show you how to predict conformational changes of your proteins using our web server

It's pretty simple and straightforward

Step one you need to provide protein structure information either by uploading protein PDB files

or by giving PDB codes and selecting chains

I will go ahead with two proteins mode first

Let me upload two protein PDB files from local computer

For protein A it's a receptor in complex for protein B it's a ligand in complex

And click this upload button

It tells me that files were successfully uploaded to the server

Step two input parameters. cut-off distance, force constant

maximum number of normal modes are three parameters

fed into cNMA framework

We have three options of

machine learning models. And you can choose in what way the job is submitted

We recommend these default values if you don't have preference

Now click this calculate button to submit the job to the server

The server is working on the job, please do not refresh page otherwise all the related files will be removed

Here we go. It's a list of all result files.You can click on the name of the file to download this file

Or you can click this download all files button to download a zip package containing all the files

We use Jmol to visualize predicted conformational changes

We calculated 20 potential vibration modes. You can check each mode here

Let's change to mode seven

The extent of the vibration increases

when the color shifting to red

while decreases

when shifting to blue

this vibrating part is

receptor. This static part is ligand. You can see the interaction between receptor and the ligand

Some animation options can be easily changed here, you can turn on and off vibration

You can show and hide vectors

You can change the frequency of vibration

or change the scale of extent

change the scale of vector length

let me zoom in so you can see it clearly

Or change the width of vectors in pixels

and also you can change the color of vectors

You can download a screenshot of current Jmol window here. We provide different formats

Then let me repeat similar process in one protein mode

This time I will input PDB codes

And select chain A and chain C

In the same way you can download files here and play with visualization here

There are some other small features worth mentioning. This part shows some statistical figures

Total number of users, total number of submitted jobs

number of all countries where visitors come from

Here we present a visitor location map

Each job has a folder on the server. If this page is closed, refreshed or

Idle for more than 10 minutes

The folder and its contents will be removed

We are working with our IT department to make this web server accessible to the public

The address will be published once it's available

Now a few years ago, I was introduced to Patacon Pisao.

It's a sick little Venezuelan mom-and-pop joint,

but their $2 tequeños are bangers.

Now I know they have two locations in the city.

One uptown and one in the LES.

So you know I'm hitting up the LES.

It's closer, and there's more stuff to do.

So these are the rules of "Operation 5 Buck Lunch."

It's got to be good, 'cause nobody

wants bad food for lunch.

No big chains, 'cause that's just too easy.

It's got to be $5 or less.

And whatever I don't use today,

rolls over into the next episode.

OK so what is your favorite food?

In this entire show, this is the first time

we've heard oysters but that's, actually, pretty solid,

is a Venezuelan appetizer.

They can be eaten for breakfast, lunch, dinner, after-hours.

It's, basically, think of cheesy bread.

Cheese wrapped in dough and fried

until it's nice and moist inside.

And cheesy inside and crunchy outside.

When you dip it into our sauces,

it's just out of this world.

- Guys, you're welcome, you're welcome.

How many tequeños can I get for five bucks?

- You can get three of them for $5.

- That's amazing.

Can I please have that?

- Of course man.

The bread is crispy,

I have some decent experiences with rosada,

but verde is my god.

If I could put this on everything in the world.

or cereal or waffles,

I would do that.

Verde sauce is my president.

Now by itself, this is good

but with the verde sauce,

It's like an aioli, close to.

Mild, it kinda oniony.
