Youtube daily report Jan 18 2019

- [Instructor] So at this point in the class,

what we want to start doing is thinking about how

we can apply the concepts we learned about

amino acids and peptides to,

considering how proteins are constructed.

So just as a reminder here,

when we're thinking about amino acids and peptides,

alright so we're talking about single amino acids,

which we know have an amine-terminus.

We go through an alpha carbon which has

an R-group attached to it.

And then we have carboxyl-terminus, so a C-terminus.

And when we have multiple amino acids that

combine together through a condensation reaction,

we end up forming a polypeptide chain.

Which,

we so we still have single amino acids.

But now instead of attached to an oxygen,

or attaching to another nitrogen through this peptide bond,

we have now a second R-group.

Another carbonyl

We attach to nitrogen through another peptide bond.

Our third R-group.

And we proceed in this manner.

Alright so when peptides form, we form these peptide bonds.

And that gives these new molecules,

these new polypeptides,

the properties of each of the

individual amino acids side-chains.

So when we talk about protein structure,

we break it up into three categories.

We have Primary structure.

We have Secondary structure.

And we have Tertiary structure.

And we abbreviate these one,

two degree,

and three degree.

Okay so our Primary structure consists of

the amino acid sequence of a protein.

Amino acid sequence.

Okay so when we think about the properties of

the primary structure of a protein,

what we're really thinking about is the functionality

that it has because of the sequence of amino acids.

So for example if we have a peptide that's alanine,

valine, lysine.

Right so the Primary structure has everything to do with

the side chains.

So here we have a CH3.

Valine we know have a CH, and these two CH3s hanging off.

The lysine we have CH2, CH2, CH2, CH2.

And then we have this NH3 hanging off down here.

Right so when we talk about Primary structure,

we are talking about the amino acid sequence

and the chemical and physical properties that are conferred

to this polypeptide chain because of the side-chains.

Okay?

When we're thinking about Secondary structure,

I'll change colors here.

When we're thinking about Secondary structure now,

what we're thinking about is how the Backbone

of a polypeptide chain can,

adopt certain structures.

Okay, and the two really big structures that

we're gonna care about are Beta-sheets,

and Alpha-helices.

Okay so these two Secondary structural elements,

or Secondary structural motifs are gonna have dedicated

videos to discuss them.

But so just in general when we're thinking about

Secondary structure, what we're really thinking about

are these,

these confirmations that fold, these structures that fold,

within the polypeptide chain.

Okay so for our Beta-sheets,

when we look at a cartoon diagram we

see them as these two-dimensional arrows.

And they're oriented as such that the arrow points from

the N-terminus to the C-terminus.

Okay so when we think about our Secondary structure,

if this line represents the polypeptide chain from the

N-terminus to the C-terminus then

whenever we see this arrow,

that means that all of the amino acids within this region of

the polypeptide chain are part of a Beta-sheet.

Okay, our Alpha-helices we represent as cylinders.

Okay and again these are always gonna be oriented from

the N-terminus to the C-terminus.

Okay so within the polypeptide chain,

if we have a cylinder this means that these amino acids,

this region of amino acids are part of an Alpha-helix.

Okay so if we continue on here.

Alright so these amino acids will be part of the Beta-sheet.

These amino acids will be part of the Beta-sheet.

These amino acids will be part of an Alpha-helix.

Alright so this is a cylinder.

Okay and then we finish up over here at our C-terminus.

So we can show these diagrams of the polypeptide backbone,

with these cartoons or these images drawn in,

that represent Secondary structural elements.

And we always show Alpha-helices as cylinders,

and Beta-sheets as arrows.

Okay?

So Tertiary structure then,

we'll start if off down here again.

So Tertiary structure is thinking about the interactions

between the Secondary structural elements.

So it's how the Secondary structure

compacts itself to form the true three-dimensional

structure of the protein.

Alright and this drawing that I'm gonna give here,

that I'm gonna show you here,

probably doesn't do justice to what's really happening,

but what I'm gonna try to do is kind of

show how these Secondary structural elements might combine.

Okay so I'm gonna go ahead and label this as Beta-sheet one,

Beta-sheet two, and Beta-sheet three.

Right so the idea here is that these three Beta-sheets,

can combine into one full sheet.

Alright if here we show the first one,

from here we kind of go on and,

we have an Alpha-helix,

I'm gonna make a curve here.

We then have an Alpha-helix.

Okay and then we then transition into the second Beta-sheet.

And then we transition into the third.

And then finally we would go on to our last helix here,

which maybe ends up pointing this way.

Okay and then off to our C-terminus.

So the idea here is when we're thinking about

Tertiary structure what we're thinking about is how

these individual Secondary structural elements

interact with each other.

And as we're gonna talk about in

I guess towards the end of this unit,

is that the interactions between

Secondary structure elements,

really boil down to chemistry between the side-chains.

So Tertiary structure boils down to side-chain interactions.

Where Secondary structure up here really boils down to

interactions between the backbone,

of within the backbone of the polypeptide chain.

Okay so,

the next few lectures are gonna

focus a lot more on Secondary structure.

And then we'll kinda finish up with thinking about how the

Beta-sheets and the Alpha-helices can interact in a way to

form a compact protein structure that

can have a very specific function.

- [Instructor] Earlier in this unit,

we talked about the three levels of protein structure.

Primary structure is all about the amino acid side chains

and the order of amino acids within a polypeptide chain.

The secondary structure are alpha helices

and beta sheets.

And these, we learned, were held together

by hydrogen bonding within the backbone.

So B.B. here for backbone.

And the third level, which is really we're gonna kind

of focus on in this video is our tertiary structure.

And this is the three dimensional structure of the protein

which involves interactions

between secondary structural elements.

So it involves the interaction between alpha helices

and beta sheets.

And since the backbone of the polypeptide chain

is tied up as secondary structure, right,

so the backbone is no longer reactive,

so the forces that stabilize this three dimensional

structure must be the side chains of our amino acids

within the polypeptide chain.

So that's what we really wanna focus in on today.

Just to kind of give this, I guess, a little bit

of a pictorial view, if we focus in on our,

this cartoon diagram of a polypeptide chain we have up here,

we see that if we start at the end terminus,

the first secondary structure element we run into

is a beta sheet and the second one

also is gonna to be part of a beta sheet.

The third and fourth are alpha helices

and the fifth and sixth here

are also gonna be part of beta sheets.

So if we think about how these secondary structural elements

might come together to form a tertiary structure.

So we start with the end terminus,

we first proceed through our first strand here

that will be part of a beta sheet,

and we see that the second secondary structural features

also are gonna be part of a beta sheet,

so maybe we form this anti-parallel beta sheet here, right.

As we talked about, this is gonna be stabilized

through this hydrogen bonding

between the carboniles and the nitrogen

within the polypeptide backbone.

The next secondary structural feature we see

is this alpha helix, followed by another alpha helix.

So perhaps this alpha helix could maybe sit on top

of our beta sheet here, right.

So we have this interaction then between our beta sheet

and our alpha helix that must be stabilized

by side chain chemistry.

The next secondary structural feature is another alpha helix

and maybe it's sitting out over here.

And then, finally, we have our last beta sheet,

which perhaps it's sitting something like this, right.

So I'm making up this structure as I go,

but what I'm hoping that makes sense to you

is that when this structure compacts

into the structure that we know it needs to

to give it it's function,

there has to be interactions

between these secondary structural elements that exist

in the polypeptide backbone.

And these need to be held together

by interactions with side chains.

So then, without going into a lot of detail

about every single intermolecular force that can exist

between side chains, let's instead kind of focus

on the overall big picture of tertiary protein structure.

So what ends up happening,

the reason these proteins fold into the structures they do

is because they're trying to take all

of the hydrophobic amino acids,

all the hydrophobic side chains and shove them together

into the core of the protein.

So this, what I'm drawing here, is a hydrophobic core.

So it's all of the non-polar side chains being shoved

into the middle, so a non-polar core.

And then decorating outside then are polar amino acids

that can interact well with water.

So we have this, I guess, this layer on the outside

of amino acid side chains that can interact with water.

So we have a hydrophobic core

and we have a hydrophilic outside.

And this type of idea, I hope, rings a bell

because we've seen biochemical molecules adopt this type

of structure before.

Remember we talked about the lipid bilayers,

we talked about how we have our polar head groups

arranged such that our non-polar hydrocarbon tails

are all kind of packed into the middle, right.

So we have our non-polar tails all packed into the core.

So the lipid bilayer forms

in a very similar way that proteins fold,

we are keeping the hydrophobic parts away from water.

And, as it turns out, this is due to something known

as the hydrophobic effect,

which is very much driven by entropy and not enthalpy.

This is a little bit surprising

but it boils down to water molecules being displaced.

We will learn more about this

in an activity that we do in class.

Torte Kitchen Tips #1 - 5 Things Friday - Duration: 10:26.

Happy Friday! It's that time of the week again. It's time for a 5 Things Friday

and this week I am sharing five Torte tips. So are you a home baker? Are you a

wannabe chef? Do you just like to dabble in the kitchen? Then this is the week for

you. Five tips, get ready.

One of the best things about writing the Bakeshop

Mysteries for me has been my research. I have had the honor and privilege of

getting to spend lots of time with professional chefs and pastry chefs. They

have taught me so many tips and tricks that I try to share with you in the

series and these are some of my quick and easy top 5 tips. So let's get started.

Number one: Do you love coffee as much as I do? I have a serious coffee addiction.

My husband, if he doesn't get up in time in the morning, I will have already

drained the coffee pot. He's never happy about that. One of my favorite tips,

though, for keeping your coffee piping hot is so simple and easy it is this:

Take your favorite coffee mug while your pot of coffee is brewing in the morning

and pour in one cup of water. Easy. You're going to take this and put it in the

microwave for 30 seconds to 1 minute. So now my mug is toasty warm. In fact, I

don't want to let go of it at the moment. It's nice and hot. I am very

conscientious about conservation and caring for our planet so I would

recommend please don't dump this water back down the sink. I just I was pour it

back in, it's hot, but I'll let that cool and then I'll use it to water my plant.

Then I pour myself a steaming hot cup of Jules' favoriteBakeshop Blend and tada

my coffee is steaming and it will stay warm so much longer by starting with a

hot mug. Number two: Are you like me,

do you love anything Italian? Especially with fresh herbs like basil? I have this

basil here and it's so aromatic I just want to take a little chunk off. Well

prior to writing the Bakeshop series I had no idea how to actually cut basil. I

would go to an Italian restaurant and the basil would be in these like

beautiful little strips on top of my pasta and when I tried it at home it's

like all mushy and gross. Well, one tip, super easy tip that I learned

from a professional chef is how to chiffonade. I hope I'm saying that right.

Professional chefs you can stop me. First and foremost, as with any good mystery

you need a good knife. And as Jules always says, "Safety first,"

so my knife has a protective sleeve on it. Okay, so the first thing I'm gonna do

is cut off the stems and discard them. And then what I'm going to do is I'm gonna

take four, maybe five, but let's start with four, about the same size pieces of

basil. Oh my gosh, if you guys could smell what I'm smelling right now. I'm gonna

layer them and then I'm gonna roll them up nice and tightly and then I'm gonna

start from the end and I'm simply going to go slice, slice, slice. Jules, if she were

teaching Sterling, her sous chef, would say, "Be careful. Watch your thumb. Don't cut

off your thumb in the process." And let's make sure there are no potential murder

victims running around Torte, but it's that easy. And then, look, you have this

beautiful, wonderful sliced basil to sprinkle on top of your pasta or to add

to any of your favorite dishes as a garnish. Okay, we are on to Torte tip

number three. This is for you bakers out there. This is one of my favorite and

easiest tips and I can't believe it took me so long to learn it. And that is

grating your butter. Oh my gosh, this is going to be such a lifesaver and

a game changer when it comes to flaky biscuits

or making pie dough. Anything where you want those delicious layers. Instead

of like chopping butter or even mixing it up, I learned from pastry chefs one of

their favorite tips is to shred butter. Brrr...it's kind of chilly. That's all part

of the fun. Okay, so I had had my butter frozen for 30 minutes, I have a bowl, and

just a basic grater, and I am simply going to shred it just like you would

cheese or carrots or anything else you're working on. And this simple

technique is going to add such new life and fluffiness to any of your bakes. I

mean, come on, look at how beautiful that is.

Grated butter. Who knew? It's gonna change your life.

Number four: Whip it good. That is what Bethany would say. In fact, I think in

maybe one of the books she has a t-shirt that says, "Whip it good." That may be

coming to an Etsy shop near you, so stay tuned. Okay, here is such an important

Torte tip for your frosting, your buttercream, or whatever kind of frosting

you're making, really. Frosting takes time and a good, good whipping.

I learned this a few years ago from an incredibly talented pastry chef who I

know and that is that the longer you whip your buttercream the lighter, the

fluffier, the smoother it's going to be. Prior to learning that tip my

buttercream was always kind of like full of lumps of butter, and it didn't

spread very well, and I learned that you have to whip for at least three to five

minutes. Maybe longer. So let's check this out. Here's what my

buttercream used to look like. I mean, come on, it's just lumpy.

There's no smoothness. It doesn't even really look like frosting. It looks much

more like the start of cookie dough or something. I use my gorgeous KitchenAid

mixer for everything and it does the heavy lifting on this. I set it too high

and I whip it until it's lighter than air. Okay, so I

have spared you the real-time watching of the buttercream turn into its

luscious, beautifulness, but can you see the difference. This is light, airy, so

spreadable. Oh my gosh, I love it. Thank you

Torte tip number four. Whip it good. Whip your buttercream! Oh, I gotta eat it.

All right, last but not least, Torte tip number five.

Jules shares this tip, I can't remember what book, but I think when she's making

a Southwestern Chicken Soup so you'll have to remind me where she makes

tortilla soup. It could be Caught Bread Handed, maybe, I don't know. You'll

tell me, right? Anyway, I'm back with my favorite kitchen tool, my KitchenAid

mixer, which is like my baby. My husband bought me a KitchenAid mixer many, many

years ago and I don't know how I lived without it. I'm not getting paid to say

this! I just absolutely love it. This one is my cherry red and now I feel like I

need a matching Torte blue. Hint, hint. Anyway,

especially with so much baking right? 31 days of Pie? I mean, this mixer, it was put

to the test. In any event, my last tip actually comes from one of my favorite

online chefs. My husband is a really good cook, too. I tend to be the baker in our

house and he is the cook. And he is also just like so great about finding

interesting people to watch online and he hooked me on to Damn Delicious. Chungah,

who I learned this tip from and I thought it's so easy and it's so hand-saving

and that is this -- when you are making chicken that needs to be shredded

you're going to need a paddle attachment for this one and what you'll do is you

will boil the chicken until it's nice and soft and tender. Go ahead and strain

the water and then you're going to put it straight into the KitchenAid mixer

and use the paddle attachment to shred it. It's magic. Okay, so

my chicken is piping hot, it's steaming. I would not touch it. I have just drained

it and, usually, this is where I would be burning my fingers in the process of

trying to shred it. But instead, what I'm gonna do is I'm gonna pop open my mixer,

dump in the chicken, and shred. This is a process that you're gonna want to watch

closely. It's going to give you beautifully shredded, ready-to-eat chicken for your

soup, your pasta, your rice bowl. Whatever you're in the mood for. You can take it

up one notch, and you're done.

WHY YOU NEED TO BUILD AN EMAIL LIST IN 2019!! - Duration: 12:51.

Today I'm gonna break down why it's absolutely crucial that you build an email list for your business now guys

It doesn't matter what business you're in even if you're not a digital marketer online market or anything like that

You still need to build an email list for your business?

And yes, it is 2019 and a lot of people are saying the email marketing is dead

But I want to break down today five different reasons of why you should still build an email list

Even though it is 2019 even though people say it's dead which is not completely dead guys. It's dying a little bit slowing down

Definitely, but here's the 5 reasons why you should still be focusing most of your marketing efforts on building an email list

And for those you guys wondering like well Jason

How do I actually build an email list that's gonna be a video for another day

but basically all you do is run a Facebook ad to a

landing page or a lead capture page basically the same thing and then you get that person to

opt-in with their name phone number email address

So on that landing page, you're basically offering something of value in exchange for their contact information from there

You're able to go through and have an email follow-up a text message follow-up a phone call fall

Depending on what business you're in and so that's the simple thing Facebook ad Instagram Adam

whatever type of ad whatever you're using to a

Landing page and you can grab their name phone email just their email just their email phone their name and their email

Really?

It depends on what you want to collect but just know the more information you're going through and collecting the lower your conversion rates now

Guys, this video is not about how to go through and build an email list

There's a million videos out there online

But this is why you should still be building an email list in 2019. And the first reason why is because

Email out of all the audiences. I've got a facebook messenger list. I've got a text message list

I've got an email list. I've got all these different platforms. It is the number one

Revenue generating source when it comes to going through and blasting out a marketing message

Blasting out a promo or anything else which is kind of ironic because if you look at the open rates and the click-through rates on

Facebook Messenger and even on text messaging you're gonna have like a 95 percent or more the people that actually receive those message

messages that actually open those and then you're gonna have a lot higher click-through rate a lot more people clicking on those as

opposed to an email whereas with email

usually it's gonna be about 15 to 25 percent of the people that you sent it to are actually opening those emails and

Then even fewer obviously that are actually clicking on the links inside of your emails

However with every single promo that I have done in 2018 email by far

Outperforms anything on facebook Messenger and I haven't really messed with text messaging just because text seems sometimes a little bit too intimate

And it can kind of annoy people even Facebook messenger can get to that point a little bit

So you just kind of got to know what is socially acceptable and what's not and honestly

I stopped sending any marketing promo materials via Facebook messenger just because I

Just figured it doesn't work that well and is a lot better for just educational content

Let him know of a new video like this video

But the reason why I think that email is so much better when it comes to actually going through and generating sales for your business

is because when someone is going to their email

They're almost looking at it as like a to-do item to do this a checklist

something that they just got to go through and like

Hit every single item

And so when they're going through and clicking on your email if they click not obviously there's only going to be 15 to 25% They

Click, but the people they go through and click

they're focusing their attention on that email for that given time and they're treated almost like a

checklist on a to-do item that they have and so they're giving a lot more thought a lot more attention to

That email and what you have to say as opposed to a Facebook message

Yes, you get a little notification

Yes

everyone clicks that so that the notification goes

away

and they open it just because they don't want to have that red thing there because if you do

You're just gonna go crazy. Just like oh man, I gotta click that just so it goes away

At least that's how I am. I don't know about you guys and then because they're not in that like to-do list checklist mentality

They're not really giving that message that you sent the same amount of thought as your email that you sent

So a lot of times all they're really doing is just clicking on it

Just that notification go away and sometimes they might read it. Yeah, they might click on it

There's a lot of people actually click on it. But the thing is is usually they're not in the right mental state

So number one guys

Email is still the biggest revenue

generating source out of all of the different audiences all the different lists that I have in my business number two reason why you should

Be building email is still in 2019

Even though some people say that email marketing is completely dead

But it's not is that you can leverage that to build other audiences to help you grow your business

So what I've done and you guys can see right here on my youtube channel

And obviously I haven't grown crazy big in the last year

so but I was able to go over to over 20,000 subscribers here on YouTube and a large part of that is because

Anytime I launched a new video that obviously pertain to what my list is about

Then I would go through and send an email to those people and say hey by the way, subscribe to my youtube channel

so you're able to go through and build other audiences and it's the same way that I've gone through and built my

Facebook group to over 7,000 members inside of that

I haven't really focused on building that for probably about the last eight to nine months

and so that probably would be a lot bigger if I would keep on focusing on that and I've also been able to build my

facebook Messenger lists to about

16,000 people and it's all by simply

leveraging the email marketing list and as you guys know today that

Audiences are basically king

If you have an audience, then you can pretty much push whatever product behind that audience

obviously if they know I can trust you and obviously if that product makes sense to that list and

You're just gonna have that much easier of a time getting started with business

Whereas somebody that starts out in business like me a few years ago where I've got this product

I've got this service, but I have no audience. No lists nothing

It's gonna be a lot harder for that person to ramp up and get started

So if you have that audience

You've got the email list you're able to leverage and build other lists like Facebook messenger a text message list

You can build a YouTube channel

you can build a Facebook group and you can just go through and

Leverage this to go and build a lot of other community

And even an Instagram following which I don't focus on Instagram

but a lot of people love focus on Instagram so you can use that to get a lot of followers over there on Instagram to

Be in front of your ideal target

Audience now number three and if you guys have been following me my channel here for a little while

You know, I love Facebook ads I talk a lot about Facebook ads

I invest a lot of money into Facebook ads and if you have an email list

If you have an email list of let's say 100 or 500 thousand

Whatever number of people what you can do is upload that email list into Facebook and say hey

This is the custom audience and I just want my ads to show to these exact people

So now we're starting to have this like on the president's exposure where we've got an email list

So we're sending emails maybe and then we're going and we're saying hey Facebook. These are the people on my email list

So these are the people I want to show my facebook ads to so you're now showing them Facebook Ads

Instagram ads you're in their email INBOX and then what you can also do from that custom audience once you upload that

Existing email list is you can create a look-alike audience of the people on that email list

So basically want to look like audiences is saying hey Facebook

I want you to go find people that look just like this person. There may be the same age the same

interest the same

demographic say I'd like there a lot of

Similarities to your existing clientele and then you're able to go and have a bigger audience that you can run your Facebook ads to

Ultimately building your email list and just having everything grow all together

Now the fourth reason why it's very similar to the Facebook look like audiences

But if you've ever run Google Ads Google AdWords is they have something called

Similar audiences, which is literally the same exact thing as Facebook look-alike audiences

It's just Google's version of the look like audience is called

Similar audiences and you can also upload that contact information. So all those emails and have that custom audience

So now you literally have this omnipresent exposure online. You are sending emails to this person

Okay, you have your Facebook Ads showing to these people you have your Instagram ads you're hitting up up on YouTube

You're hitting them up on the Google search in all these different places even on other blogs

They're visiting and so literally guys this is the core

this is like the heartbeat to go in and having all this other stuff around it work and all your

Retargeting and then not to mention with those custom audiences inside of Google AdWords as well

You can create the similar audiences which obviously give you a lot bigger market to go through and advertise to now the fifth

reason, I would highly recommend still going through and building your email list in 2019 is

Because it's so easy to go through and test new offers new ideas new products new services new met

whatever a hundred percent for free

So let's say you go through and you get an idea for some new product or some new offering they wanted like oh I want

To go through and see if people would be interested in this

Well before you even make the product you can one go ask your email list or if you made the product you can go through

And sell it to your email list blast out one email or a couple of emails and see how the conversions go

and so or like if you're let's say gonna go do a

Webinar or a sales video or something like that you have this list of people that instead of having to go through and pay money

For Facebook, Ads Google Ads or anything like that

You send one email out to this email list

whether it's a thousand people ten thousand twenty thousand people and you're able to instantly get feedback on your messaging how the

actual offer is gonna convert and the price points a lot of

Information before you actually take that offer and that product that idea to cold traffic

so any new product or service you're gonna go through and

Mark it if you have an email list more than likely they're gonna know who you are

They're gonna hopefully like you and hopefully trust you and so those conversion rates are gonna perform

Extremely well as opposed to cold traffic so you're able to go through and if you have crazy good numbers crazy good conversion rates

with your warm audience with that email list then you know

Once you take it to that cold market and start running Facebook ads or Google Ads at scale

You know, you're gonna have a pretty good chance of having a winner

Whereas if you go through and you launch it to this email list

Which remember it's a hundred percent free because all you got to do is blast out a couple of emails to test this offer

and it performs kind of mediocre or

Performs at a level that you would be fine with that cold traffic
